"Meatballs" - "It just doesn't matter" scene
"School of Rock" - "Stick it to the man" scene
"Tommy Boy" - "...the other guys break pads" scene
"Signs" "Do you believe there is someone to protect us?" scene
"Before Sunset" - The talk while driving to Julie Delpy's apartment
"Once" - Driving in the car to listen to the record
I'm sure you can all come up with better scenes than I.

That's all I can think of at the time.
I think every great movie has at least one memorable scene that sets it apart. The most memorable have a million such scenes (like star wars). You could almost rank movies by how memorable their best scenes are. A few favorites:
-the flying bike scene from E.T.
-the wizard scene from the wizard of oz
-breaking down the door in the shining
-apes and the bone in 2001 a space odessey
-the heat/touch/sound sensitive room from MI
-blowing up the buildings in Independence Day
-gandaf vs the balrog in Lord of the Rings
-the "happy place" scene from happy gilmore
-walking through the factory in an officer and a gentleman
-the egg scene from cool hand luke
-car chase in Bullet
-motorcyle jump in the great escape
-hedgerow scene from the longest day
-the wedding scene from the godfather
-the crazy wheel scene from midnight run
-the trip to the cemetary in the magnificent seven
